2 INTRODUCTION Lavandula Angustifolia L., is an important aromatic, ornamental and essential oil producing plant. It is commonly known as Lavender belongs to the family Lamiaceae cultivated in the Mediterranean areas. The oil is used in perfumery, cosmetics, flavoring, detergents, insecticides and pharmaceutical industries. It has medicinal properties such as sedative, diuretic, diaphoretic, antiseptic carminative, spasmolytic, antidepressent and anti-rheumatic properties. The propagation by seeds is slow and also exhibits variation in growth rate, growth by stem is not only slow but its rooting ability is also poor and due to repeated vegetative propagation it leads to modification of the morphological and chemical characteristics (Minlet,1980; Panizza and Tognoni 1991). Also results in clonal degeneration and reduced life span, deterioration in vigor, low diseases resistance and tolerance to environmental stress conditions. Thus, micropropagation through axillary buds, leaves and apical buds has been proposed as a useful technique and alternative method to cope up with this problem.
3 MATERIAL AND METHODS In vitro axillary bud explants of Lavandula angustifolia L., were used as explants and were inoculated on Murashige and Skoog s basal medium (MSBM)supplemented with auxin and cytokinins for initiation and multiplication of shoots. Multiple shoots were subcultured on the same medium to obtain well developed, elongated multiple shoots. All the cultures were maintained at a temperature of 25±2 o c with photon flux density of 30-50µ µem -2 S -1 under photoperiodic regime of 16h light and 8h dark cycles.
4 Cont The in vitro raised plants with well developed roots were subjected for hardening in a potting mixture, containing peat: perlite: vermiculate (1:1:1 v/v). After 50 days of hardening, the plants were transferred to the soil. The principle component linalool both in in vitro and in vivo leaves and stem of L.angustifolia were determined by Gas Chromatography. The in vitro axillary buds were used for synthetic seed production using Sodium alginate and Calcium chloride as matrix and complexing agent for encapsulation of axillary buds of L.angustifolia.
5 Fig-1 Fig-2 RESULTS The in vitro axillary bud explant were inoculated on MS Basal Medium (Fig.1) Shoot initiation with 2-3 leaves (Fig.2), was observed after 10 days of culture on all the concentrations of growth regulators studied with varying percentage of response. (Table1,Graph1). The highest 91% on MSBM+ BAP 8.88µM +NAA2.68 µm (Fig.2). After 20 days of culture on the same medium 2-3 multiple shoots were noticed (Fig.3). Fig-3
6 Cont After 27 days of culture the number of shoots increased 3-5 (Fig.4). After 36 days of culture, the shoots were subcultured on the same medium to obtain more number of multiple shoots After 14 days of subculture,6-8 multiple shoots were noticed (Fig.5) Fig-4 Fig-5 After 28 days of subculture and multiple shoots (Fig.6) were formed. Fig-6 Fig-7 After 42 days of subculture elongated multiple shoots were noticed (Fig.7).
7 Cont v Roots. Fig-8 The Multiple shoots were inoculated on rooting medium MSBM+BAP 8.88µM +NAA 2.68µM + IBA 4.92 µm After 28 days of culture, these multiple shoots developed roots (Fig.8). Fig-9 Later these plantlets were subjected to hardening (Fig.9).

11 CHROMATOGRAM OF LINALOOL IN VITRO LEAF and STEM OF L. ANGUSTIFOLIA Leaves and stem of in vitro and in vivo of L.angustifolia were subjected to phytochemical analysis for the determination and comparison of the principle component Linalool.
12 Sodium alginate was selected for encapsulation in comparison with Carboxyl methyl cellulose which failed to complex with CaCl2, 4% concentration of sodium alginate was the best matrix and 50mM CaCl2 was found to be the best complexing agent for 40 min. exposure to obtain firm, transparent and uniform beads. Fig-10 Fig-11 The encapsulated axillary buds of L. angustifolia (Fig.10) were inoculated on regenerating medium on MSBM fortified with BAP (8.88 μμ) +NAA (2.68 μμ) in order to retrieve the plantlets. Seeds germinated after 18 days of culture with 94% of response (Fig.11) Fig-12 Fig-13 Shoot initiation with 2 4 leaves was noticed after 25 days of culture (Fig.12). with increase in the no. of shoots to 2-3 (Fig.13) after 32 days of culture

After 42 days of culture, elongated 8-12 shoots were observed (Fig.14) Fig-14 Fig-15 Healthy, elongated shoots (20-25) were observed after 56 days of culture These shoots then formed roots on the same medium after 63 days of culture (Fig.15) Fig-16 Fig-17 The well developed multiple shoots with roots (Fig.16) were subjected to hardening process (Fig. 17).

DISCUSSION It was found in the present study, that MS basal medium supplemented with BAP (8.88 μμ) and NAA (2.68 μμ) was the best medium for in vitro axillary bud initiation and shoot multiplication of L. angustifolia. Further, it was significantly superior than the other growth regulators tested. This finding does not coincide with the findings of Panizza and Tognoni (1992). However this agrees with the findings of Miguel and Maria (1996) who have reported that the micropropagation of L. latifolia is through axillary bud culture on MSBM supplemented with BAP and NAA.The present findings are in accordance with the findings of Anamaria et al., (1998) and Andrade et al., (1999) and Dias et al., (2002). Plants of lavender produced in vitro, acclimatized and planted in the field showed good quality of characteristics and were true-to-type in essential oil composition. Production of plants in vitro could help to counteract the effects of the decline phenomenon occurring in lavender.
16 Cont Micropropagation in lavender through axillary buds can be considered as a reliable method for rapid multiplication of elite clones as well as a potential useful tool for overcoming clonal problems. In the present investigation, it was found that high percentage of Linalool % percent in the in vivo leaf and stem of lavender when compared with in vitro leaf and stem (0.004 %). In the present study 4% Sodium alginate upon complexation with 50mM Calcium Chloride (CaCl 2 ) for 40 mins was found to be best hydrogel and complexing agent for encapsulation of axillary bud. Further, the firm and uniform beads so obtained resulted in retrieval of plants through in vitro germination of synseeds. The synthetic seeds of axillary buds can be stored at 4 o C in sterile water upto six months. They germinate and develop into plants when transferred to room temperature on MS basal medium fortified with BAP (8.88µΜ) and NAA (2.68 µμ) for Lavandula angustifolia.
