715001558, REV. C 1 OF 10 This procedure describes how to install the ACQUITY UltraPerformance LC System 30-cm Column Heater/Cooler (Figure 1) on an ACQUITY UPLC System. 715001558, REV. C 2 OF 10 Procedure The procedure to install the 30-cm column heater/cooler includes six subtasks: 1. Installing the adapter bracket 2. Installing the 30-cm column heater/cooler 3. Making fluidic connections 4. Making electrical connections 5. Powering on the column heaters 6. Controlling the column heater through Empower or MassLynx CAUTION: IF YOUR ACQUITY UPLC SYSTEM INCLUDES A HIGH-TEMPERATURE COLUMN HEATER WITH A REAR DSUB CABLE, MAKE SURE TO UNPLUG IT FROM THE SAMPLE MANAGER BEFORE PERFORMING THIS PROCEDURE. Installing the Adapter Bracket 1. Turn off power to the sample manager and column heater. 2. Using a T20 Torx screwdriver, remove the top screw from the right side of the sample manager, and the bottom screw from the right side of the binary solvent manager (Figure 2). 715001558, REV. C 3 OF 10 3. Mount the rear tab of the adapter bracket onto the ground stud at the rear of the sample manager (Figure 3). If the ACQUITY UPLC system includes a sample organizer: Position the bracket so that its inset area fits over the sample organizer leg (Figure 4). Insert two M4 x 16 screws with cup washers into the slots on the adapter bracket s left panel, leaving the first screw loose until both screws are inserted (Figure 6). Tighten both screws. Figure 6 - Attaching the Adapter Bracket 3. Place the blue screw caps over the M4 x 16 screws (Figures 5 & 6).

715001558, REV. C 5 OF 10 4. Add an M4 Keps nut (WAT251620) (Figure 7) to the ground stud at the rear of the sample manager (Figure 4) and tighten with a 7-mm or adjustable wrench. Insert the three M4 blue cover screws (PN 410001262) through the three side panel spacers (WAT270649) (Figure 8) and then screw them into the threaded holes in the adapter bracket s center panel (Figure 6). Figure 8 - Spacer and Cover Screw 6. Mount the 30-cm column heater/cooler onto the adapter bracket, fitting the column heater/cooler mounting holes (Figure 9) over the M4 cover screws. 715001558, REV. C 6 OF 10 Making Fluidic Connections 1. Using a 1 / 4 bit driver, attach the three black tubing clips to the front of the bracket. Load each clip into the bit driver in the orientation desired, and then snap it into the bracket. (Figure 10) Tubing clip Figure 10 - Installing the Black Tubing Clips

715001558, REV. C 7 OF 10 2. Connect the stainless steel inlet tube (PN 430001923, Figure 11) from the column to the inject valve port 6 (Figure 11). NOTE: You must install the stainless steel inlet tube in order to perform AQT testing. 3. Connect the red PEEK tubing (PN 430001919, Figure 13) from the outlet of the column to the detector. Slide the drip tray onto the bottom of the column compartment, inserting the L-tabs on each side of the drip tray into the slots in the column compartment floor (Figure 12). 5. Route the tubing to a suitable waste container. Making Electrical Connections Figure 14 - Installing the Drip Tray 1. Connect the Y-cable to the 30-cm column heater/cooler and high-temperature column heater (HTCH) (Figure 15). CAUTION: MAKE SURE POWER IS TURNED OFF WHEN CONNECTING OR DISCONNECTING THE Y-CABLE. 2. If the ACQUITY UPLC system does not include an HTCH, make the Ethernet connections: a. Place an 8-port network switch box on top of the column heater/cooler (Figure 15) and connect one end of a shielded Ethernet cable to the network switch, and the other end to the Ethernet card on the workstation. b. Connect one end of a shielded Ethernet cable to the network switch, and the other end to the Ethernet card of each of each instrument in your ACQUITY UPLC System. c. Plug the Ethernet switch box power cord into a suitable wall outlet.

Network switch box Y-cable Power cord Figure 15 - Making Electrical Connections 3. Connect one end of the power cord to the column heater/cooler and the other end to a suitable wall outlet. Operating the Column Heaters The column heaters contain an internal Ethernet switch that allows the ACQUITY UPLC system workstation to communicate with all the instruments. To make sure that the software properly recognizes all the instruments, use the correct sequence to power on the instruments. NOTE: You must power-on the HTCH, column manager, column heater/cooler, or 30-cm column heater/cooler before any of the other instruments. NOTE: For systems containing both a 30-cm column heater/cooler and an HTCH, you must power on the 30-cm column heater/cooler before powering on the HTCH. See the following guidelines for the correct power-on sequence.

715001558, REV. C 10 OF 10 To operate both a 30-cm column heater/cooler and an HTCH: 1. Power on the 30-cm column heater/cooler. 2. Power on the HTCH. To operate a 30-cm column heater/cooler alone: 1. Power on the column heater/cooler. 2. Power on the sample manager. To operate an HTCH alone: 1. Power off the 30-cm column heater/cooler. 2. Power off the sample manager. 3. Power on the sample manager. 4. Power on the HTCH. Controlling the Column Heater through Empower or MassLynx To access the instrument method parameters for the 30-cm column heater/cooler, go to the sample manager control panel in Empower or MassLynx (Figure 16).
