3 CHAPTER 1: INTRODUCTION For histological diagnoses, tissue specimens for microscopic examination have to be prepared by processing tissues through a series of given procedures. In general, preparation of tissue specimens consists of processing (fixation and dehydration of tissues), embedding, sectioning and staining. The Tissue-Tek TEC Plus Console System is used for the embedding procedure. The Tissue-Tek TEC Plus Console System consists of two consoles; one is Tissue-Tek TEC Plus Dispensing Console (for embedding tissues in paraffin) and the other is Tissue-Tek TEC Plus Cryo Console (for cooling paraffin blocks). This Operating Manual provides operating information on Tissue-Tek TEC Plus Cryo Console for cooling tissue-embedded paraffin blocks Ver. June 2003
4 [1] Notation Notation related to Safety CHAPTER 2: OPERATIONAL PRECAUTIONS In this Operating Manual the following notation is adopted for safety operation. The contents of accidents which will occur from improper handling caused by neglecting the precautions are classified as follows. Have first a well understanding of contents and then read this Manual. WARNING indicates a potential hazard which, if not avoided, could result in death or serious injury to personnel. CAUTION NOTE indicates a potential hazard which, if not avoided, could result in minor injury to personnel or damage to the instrument or other property. is a reminder or lists helpful information. [2] Safety Precautions Since improper operations with safety precautions ignored may result in personal injury or property damage, instructions herein must strictly be observed. WARNING Do not disassemble nor remodel the instrument in any case. Doing so will lead to abnormal instrument operations or cause a mechanical/electrical accident which is dangerous. WARNING Do not wet any parts of the instrument except as described in Chapter 5 Maintenance. If the instrument gets wet, an electric leakage may possibly lead to a fatal electric shock or fire accident. WARNING Do not operate keys and switches with a wet hand. Otherwise, it may cause an electric shock accident. WARNING Never plug or unplug the power cord with a wet hand. Otherwise, it may cause an electric shock accident. WARNING Hold the plug to connect or disconnect the power cord. Failure to do so may lead to a fatal electric shock or fire accident. WARNING Do not damage the power cord. Do not forcibly bend, pull, twist or bundle the power Ver. June 2003
5 cord. A damaged power cord can cause a fire or electric shock accident. If any unusual condition is found on the power cord or plug, contact Sakura local distributor or representative. CAUTION CAUTION CAUTION The power cord must be plugged into a grounded, dedicated power receptacle. If a grounding adapter is needed, it must be connected to a grounding terminal. Failure to do so may cause a fire or an electric shock accident if current leaks. Make sure that the instrument power is off before attempting daily care of the instrument. To do otherwise may cause unexpected accidents. In the event that any abnormal instrument behavior occurred, turn instrument power off and unplug the power cord. Contact Sakura local distributor or representative for technical assistance and service. CAUTION Do not peel off warning or caution labels provided on the instrument. To avoid unexpected accidents, these labels are necessary to call your attention to important information on routine handling and operations CAUTION The instrument should be maintained on a regular basis (every 6 months is recommended) to ensure proper instrument operations and service. For the detailed information on periodical maintenance contract, consult Sakura local distributor or representative. [3] Operational Precautions Explained below are the precautions to be observed for preventing instrument failures. Never use organic solvents (such as xylene) to clean the instrument. material and paint finish of the exteriors and the control panel. 6 CHAPTER 3: INSTRUMENT DESCRIPTION AND INSTALLATION [1] General Description The Tissue-Tek TEC Plus Cyro Console is a cooling unit to cool the base molds containing a processed tissue embedded in molten paraffin to make paraffin blocks. 7 <Description of each component> (1) Cooling Plate A chilled plate for preparing paraffin blocks by cooling and hardening molten paraffin in base molds containing a processed tissue each. This is also used as a storage place for keeping solid paraffin blocks prior to sectioning. A sealed compressor refrigerator is built in this console. The cooling plate can hold up to 64 base molds. The cooling temperature is selectable from the range of -10ºC to 0ºC. (2) Waste Tray When water drops are produced on the surface of the cooling plate, they are flowing along the sides of the plate to drain into this waste tray, which will prevent the bench from getting wet. <Caution Labels> Label (1) Do not put anything on top of the instrument. 8 (2) Control Panel Display [+] key [-] key Power Switch [SET] key [ENTER] key Power Indicator <Description of each component> (1) Power Switch Turns the power to the console ON or OFF. (2) Power Indicator Illuminates when the power switch is set to ON. (3) Display Indicates a temperature in two digits. When in the operating mode, the current, actual temperature of the cooling plate is displayed. When in the setting mode, the preset cooling temperature is blinking. (4) [SET] key When pressed, the operating mode changes to/from the setting mode. (5) [ENTER] key When pressed, a changed temperature setting is saved into memory. (6) Plus ([+]) key When pressed, a temperature value increases. (7) Minus ([-]) key When pressed, a temperature value decreases Ver. June 2003

10 [3] Installation (1) Environmental Factors Install the Cryo Console at the place where can meet requirements below. * A place with no fire hazard in the vicinity. * A place away from direct sunlight. * A level, solid and unshakable place. * A place where is vibration-free. * A place away from water splashes. * A place away from volatile or flammable materials. * A place away from corrosive chemicals (acids or alkalis). * A place away from any equipment that operates at a high voltage or a high current, e.g., a large-sized refrigerator, supersonic washing machine, cooling centrifugal machine, etc. * A place where can meet following environmental conditions: Ambient temperature : 10 C ~ 40 C Relative humidity : 30% ~ 85%RH (no condensate) Atmospheric pressure : 700 hpa ~ 1060 hpa (2) Installation Procedure (a) Main body When installing the Cryo Console only, Select a firm level bench capable of supporting the weight of instrument (approx. 22 kgs) with enough space (at least 50 cm in depth) to place the instrument on it. 11 When installing the TEC Plus Console System (1 Dispensing Console + 1 Cryo Console), Select a firm level bench capable of supporting the gross weight of the system including paraffin (approx. 50 kgs) with enough space to place the system on it. NOTE: * Place the Cryo Console side-by-side at the left hand of the Dispensing Console. * The Dispensing Console can be laid close against the back wall, but a space of 4 cm or more will allow the paraffin chamber lid to be fully opened. Approx. 56 cm Approx. 87 cm (b) Power cord (1) Ensure that the power switch is set to OFF. (2) Connect the power cord to the power cord connector on the rear panel. Power Cord Connector Power Cord (3) Insert the other end of the power cord into a properly grounded power receptacle (3P). (If the 3P receptacle is not available, use a grounding adapter.) Ver. 12 The power source needs to meet the following power requirements. Always plug the power cord directly from the instrument to a power source. Failure to do so could cause low supply voltage that will make the compressor unworkable. 13 CHAPTER 4: OPERATION [1] Instrument Setup (1) To start the instrument operation, set the power switch to ON. At the beginning of the operation, the display indicates the current temperature of the cooling plate. 2 0 ºC (2) Press the SET key, if necessary, to change the preset cooling temperature. The instrument will enter the setting mode and the current preset temperature will blink on the display. - 5 ºC (3) Press the plus or minus key as necessary to reach a desired temperature. Holding these keys for more than 2 seconds enables rapid scrolling. The allowable range is -10ºC to 0ºC. The initial factory setting is -5ºC. (4) When the desired value is obtained, press the ENTER key to save the selected temperature. To exit without saving the value, press the SET key again. The selected temperature is stored in memory. The display returns to indicate the current temperature of the cooling plate. (5) After the routine embedding work has been finished, make sure to turn the power switch off to stop the instrument operation Ver. June 2003
14 [2] When Power Outage Occurred * When a power outage occurred during the operation, the temperature control stops functioning and the cooling plate begins warming up. However, when the power is restored, the instrument will automatically resume the operation. * To protect the compressor, it takes 4 minutes to restart the operation after the power was restored. [3] When an Error Occurred * When an error occurred on the sensors or the instrument memory, a corresponding error code is displayed with blinking. * At the same time, the buzzer starts sounding to alert of the occurrence of an error. Pressing the ENTER key can stop the buzzer. * The error code indicated on the display cannot be disappeared until the cause of that error is eliminated. * When an error occurred, contact the Sakura distributor or representative for technical information and service. 15 [1] Care of Instrument (1) Daily Precautions CHAPTER 5: MAINTENANCE Do not place anything on the top of the instrument. It will interfere with the proper ventilation. When any reagent accidentally spills on the instrument, rapidly wipe it out using a soft cloth. Make sure to unplug the power cord when the instrument is not in use for a long period of time. (2) Cleaning Wipe clean the cooling plate with a dust-free cloth or paper to remove stains and water drops from the plate surface. Use care not to damage the surface of the cooling plate. If it is damaged, water drops may corrode the coating of the surface to cause the peeling. Remove spilled paraffin from exterior panels using the scraper. Do not wipe the exteriors using a solvent like xylene to avoid damage of the paint finish. Draw the waste trays toward the front and wipe them using a cloth or paper to remove drained water. [2] Troubleshooting Problem Power indicator does not light up. Instrument does not operate. Cooling plate does not cool. Check to make sure: Power switch is set to ON. Power cord is firmly connected to the instrument and power receptacle. Power receptacle is live. Power switch is set to ON. Power voltage is within the 10% tolerance of the instrument rated voltage. Power cord is not extended using a different cord. Temperature was preset in a correct manner. Preset temperature is set at a proper value. If the problem cannot be resolved after checking the above items or the problem you are experiencing is beyond the scope of the above, contact the Sakura distributor or representative Ver. June 2003
