2 McGaugh Hall Insectary Renovations Summary description: Provisional requirements are provided here for a self-contained, secure insectary with working space for laboratory personnel and three modular units for rearing insects. All space is minimally BSL2 (ACL2) with the option for BSL2 + certification. This insectary provides adult rearing space, increase working space for mosquito technical staff, provide BSL2 + facility for rearing mosquitoes infected with malaria parasites and some viruses, and provide a safe and secure working environment for mosquito research. Structural features: (Please refer to the accompanying draft sketch of the facility). Gas closet (#1): The gas closet has mounts and hardware for four (4) large CO2 cylinders. This allows changing of the cylinders without personnel having to enter the room. The cylinders connect to gas piping that delivers the gas to positions indicated in Rooms 3-6. The room also could serve to store a small amount of insectary-related supplies. Access foyer (#2): The foyer provides controlled access to the insectary, serves as a gowning and degowning space for the insectary personnel, provides storage for personnel clothing and personal items, and provides two barriers to possible escape of mosquitoes. The room should have: -Keycard access for the outer door -Lockers for six people -Light switch for red light outside door to let people know when BSL2 + conditions are to be observed. Working room (#3): The purpose of the working room is to provide space for the care and maintenance of the insects in the colonies, some experimental work, limited cleaning of insectary materials and limited storage of insectary supplies (controlled substances, etc.). The room should have -A large sink with H&CRW and removable (cleanable) screen water traps -DI water -vent traps (screens) on all HVAC apertures -AC power on all walls, -high and low benches, cabinets and shelves (placement along all walls, details to be furnished following discussion) -Screen or other enclosures to prevent insects from entering light fixtures -CO2 piped in from Room #1 (number of valves and location to be determined) - house gas/vacuum lines (2 locations ) -Internet access 1

3 -Phone jack -water-resistant floor treatment, lightly gritted to prevent slipping when wet. -floors to have gentle slope to center of room to pool spilled water and make it easy to remove Insectary chambers (#4 and #5): These chambers provide secure space for maintaining infected and uninfected adult mosquitoes. The rooms must have independent controls for heating/cooling, lighting and humidity. Each of these is accessed by a swinging door that seals completely. Specifications for each room: Controlled temperature: Chamber temperature range: 20 o C to 40 o C. Chamber temperature control sensor: ±0.2 o C. Chamber temperature uniformity: ±0.5 o 25 o C (within working area, away from direct airflow and 6 away from all walls and floor). Exceptionally accurate NIST-traveable platinum RTD control sensor included. Controlled humidity: Chamber-relative humidity range: ambient to 90% throughout temperature range. Chamber-relative humidity control at sensor: ±0.5% 25 o C and 75% RH. Chamber-relative humidity uniformity: ±1% 30 o C and 75% (within working area, away from direct airflow and 6 away from all walls and floor). Controlled lighting: Chamber lighting: adjustable diurnal dimming incandescent lighting. Fluorescent lighting accessed by simple external off-on switch. Microprocessor controller in control console will measure light level inside chamber and be able to adjust to desired level. Chamber lighting will also be userprogrammable to automatically ramp to and from preset light levels (at any rate) for close replication of natural diurnal lighting. Microprocessor will read out measured light level as well as the desired level. Lighting to be sealed to prevent insect escape. (Condensing unit: top-mounted, air-cooled 1 horsepower condenser. Evaporator (s): HeatCraft, manufactured from stainless steel for maximum corrosion resistance. Unit sized for maximum flow volume and minimum air speed/noise level. Unit features resistance to acids generated in rearing of floor and low number of fins per inch for easy cleaning and resistance to blockage.) Exterior size: TO BE DETERMINED. Panel construction: 4 thickness, foamed in place polyurethane, 30 R-value. Panel surface finish, interior: white stucco-patterned mold-resistant aluminum. 2

Fuji PID/fuzzy Logic microprocessor for temperature, lighting and humidity control. Dual display of actual and set-point temperature, lighting, and humidity. Solid-state control relays wherever possible. High/low temperature and humidity alarms settable in 0.1 o C and 0.1%RH increments with: automatic cut-offs of equipment during high or low alarm, dry contact remote output. Electrical requirements: three circuits 20 amps/120 volts/60 hz each on their own circuit breaker with disconnects at unit. -Screening over all ducts and apertures to prevent insect escape. -Small sink with H&CRW and removable (cleanable) screen water traps -DI water -CO2 piped in from Room #1 Insectary chamber (#6): This chamber provides secure space for rearing sub-adult stages of mosquitoes. The room must have independent controls for heating/cooling, and lighting. A double hanging plastic curtain instead of a door is used for access. Specifications for the room: Controlled temperature: Chamber temperature range: 20 o C to 40 o C. Chamber temperature control sensor: ±0.2 o C. Chamber temperature uniformity: ±0.5 o 25 o C (within working area, away from direct airflow and 6 away from all walls and floor).
