3 1.0 DESCRIPTION The Thermo Savat SC110 SpeedVac ad SC110A/SC210A SpeedVac Plus cetrifugal vacuum cocetrators rapidly evaporate liquids to cocetrate or dry solutes, aalytes, ad residues i aqueous ad orgaic solvets. Vacuum, applied to the sample chamber from a exteral source, promotes solvet evaporatio. As the rotor i the cocetrator spis, cetrifugal force keeps samples from foamig or bumpig, prevetig sample loss ad resultig i superior aalyte recovery. Typical applicatios iclude cocetratig or dryig oligoucleotides, ethaol precipitates of DNA, hydrolysates of proteis, chromatography fractios, solid phase eluats, liquid/liquid extracts, ad samples for aalysis by HPLC, GC/MS, immuoassay, ad bioassay. SpeedVac cocetrators are also used i orgaic chemistry residue aalysis, such as pesticide ad drug testig. The SpeedVac SC110 model icludes a built-i maual bleeder valve. This istructio maual cotais a explicit operatig sequece (see sectio 3., SPEEDVAC SC110 OPERATION) that the user must follow to esure vacuum is applied to the sample chamber oly whe the samples are spiig. The SpeedVac Plus SC110A/SC210A models itegrate a automatic bleeder valve that esures applicatio of vacuum at the appropriate time, without user itervetio. All SpeedVac cocetrators iclude a chamber heater to effectively couteract evaporative coolig ad accelerate dryig rate. The operator ca select low chamber at ambiet temperature), medium (chamber at 3 C), or high (chamber at 65 C). For eve shorter dryig times, a optioal Radiat Cover further hastes dryig rate by addig eergy to the sample chamber. Thermo Savat offers a wide variety of rotors for the SpeedVac cocetrators. They are desiged to accommodate microtubes, bottles, glass, plastic tubes ad eve microtiter plates. The SC110/SC110A uits hold up to 0 x 1.5ml sample tubes while the larger SC210A holds up to 200 x 1.5ml tubes. A lie of large-volume (up to 500 ml), multi-sample rotors hold Savat s recovery flasks or pearshaped flasks with receptacle eds. 2.0 INSTALLATION Receivig. Ispect the shippig carto upo receipt. If the carto is damaged i ay way, do ot accept delivery. Call Thermo Savat at , , or (Fax)

4 Upackig. Carefully remove the istrumet from the shippig carto. Compare the packig list to the box cotets. If there is a discrepacy, call Thermo Savat at , , or (Fax) Ispectio. Ispect the uit for ay damage that may have occurred durig shipmet. Should there be ay damage, report it to the carrier ad cotact Thermo Savat immediately. Make sure the carrier ispects the damage ad leaves a ispectio report. Register ay claims for shippig damage agaist the carrier or his aget. Save the shippig carto i the evet a retur is ecessary. Set-up. To assure safe operatio ad best results, read this maual i its etirety before operatig the istrumet. 2.1 SITE PREPARATION Istall the SpeedVac o a clea, dry, level, stable surface withi ft (1.2 m) of a compatible electrical outlet. Place the uit i a coveiet locatio with access to a vacuum source. Typically, the SpeedVac Cocetrator sits o a coutertop or o the mobile Thermo Savat CC0/DX Deluxe Coveiece Cart. Overhead clearace, roughly equal to the uit s depth, is required for raisig the cover. WARNING: Before coectig the SpeedVac Cocetrator to a outlet, make certai that voltage, frequecy, ad amperage match the requiremets idicated o the label o the rear of the istrumet: SC110/SC110A: 0 VAC / 60 Hz, A or 230 VAC / 50 Hz, 2 A; SC210A: 0 VAC / 60 Hz, A or 230 VAC /50 Hz, A If there are ay questios, please cosult a electricia. Note: To maually ope the lid prior to applyig power, use the iterlock over-ride tool. This tool may also be used to ope the uit durig a power outage. Isert the tool at a upward agle (approximately 5 ) ito the small hole i the upper right-had pael of the SpeedVac cocetrator. While liftig up o the lid, pivot the tool hadle upward, creatig a dowward movemet withi the SpeedVac. This maually disegages the lockig mechaism. 2.2 PREPARING CONCENTRATOR FOR USE 1. Make sure the ON/OFF switch o the frot of the SpeedVac is i the OFF positio. Coect power cord to istrumet ad plug ito appropriate outlet. The safety lid lockig mechaism o SC110/SC110A- 20 V ad SC210A 0 V/20 V uits will automatically disegage. 2. Lift the lid ad clea the chamber of ay packig material or foreig items that may be preset from shippig. 3

5 3. Lightly coat the drive shaft ad rotor ceter with vacuum grease. This protects these surfaces from corrosive vapors. Do ot lubricate the cover O-rig.. Attach the cocetrator to a vacuum system (Refrigerated Vapor Trap plus Vacuum Pump) by coectig the 0.5 i (1.27 cm) O.D. hose fittig at the rear of the SpeedVac Cocetrator to the vacuum system. Coat fittig with a thi film of vacuum grease ad thread vacuum tubig through the hole i the right side pael of the cocetrator. Without twistig, carefully push vacuum tubig straight o over fittig. Note: Read the recofiguratio sectio of this maual (see APPENDIX 2, page 11) before discoectig or replacig vacuum hose. Protectig the vacuum pump: A vacuum pump experieces loss of efficiecy ad ultimate damage if evaporated solvets from samples are allowed to eter. To prevet damage ad exted pump life, a refrigerated vapor trap that codeses ad preferably freezes solvet vapors must be placed i-lie betwee the SpeedVac cocetrator ad vacuum pump. A complete lie of vacuum pumps ad refrigerated vapor traps is available from Thermo Savat. A chemical trap, placed i-lie betwee the refrigerated vapor trap ad the vacuum pump, further protects the pump from ay vapors pulled through the trap. Thermo Savat s SCT0 Chemical Trap may be used with the DC0A cartridge to adsorb aqueous vapors ad eutralize corrosive acid vapors, or with the DC0R cartridge to adsorb radioactivity ad volatile orgaic vapors. If ucertai of the optimal setup, call Thermo Savat Techical Services at , , or (Fax) for additioal iformatio. 3.0 OPERATION The SpeedVac or SpeedVac Plus cocetrator is a importat compoet i a sample dryig/cocetratio system that should also iclude vacuum gauge, refrigerated vapor trap, chemical trap, ad vacuum pump, ad may cotai other equipmet. Please refer to operatig istructios of each compoet for details o use.

6 3.1 CONTROL PANEL 3.2 ROTORS Two switches o the istrumet frot pael gover operatio of the SpeedVac SC110/SC110A/SC210A cocetrators: The CONCENTRATOR switch cotrols rotatio (ON/OFF). The DRYING RATE switch regulates sample chamber temperature: HIGH 65 C MEDIUM 3 C LOW ambiet Idicator lights illumiate whe a fuctio is switched o. A wide variety of SpeedVac rotors is available to suit every sample dryig applicatio. CAUTION: Oly these rotor series should be used i the SpeedVac. DO NOT use rotors other tha Thermo Savat rotors, i the SpeedVac Cocetrator eve if they seem to fit the istrumet. CAUTION: Always balace rotor loads. A ubalaced rotor causes vibratio that will wear out the bearigs ad may seriously damage the SpeedVac. Load the rotor uiformly. There eed ot be a tube i each place, but the tubes must be evely spaced aroud the rotor. Whe usig a rotor with alumium tube holders, isert tube holders symmetrically. Whe dryig differet types of sample i the same dryig ru, distribute each type symmetrically so that their differet dryig rates do ot itroduce precessio ito the rotor s rotatio. Always select a rotor ito which the sample tubes fit sugly. To istall or chage a rotor: 1. Switch the CONCENTRATOR to the OFF positio. 2. Ope the lid, uscrew the rotor hold-dow kob ad remove the existig rotor (if ay). 3. Select desired replacemet rotor (see rotor chart o page 9).. Alig the otches i the rotor base with the drive pi of the motor shaft ad place the rotor o the shaft beig sure that the pis egage the otches. 5. Had-tighte the rotor hold-dow kob util firmly seated. 5

7 3.3 PREHEATING Evaporative sample coolig that occurs uder vacuum actually decreases sample dryig rate. The sample chamber heater couteracts this effect to accelerate dryig rate. If desired, operate the DRYING RATE switch at least 15 miutes before startig the cocetratio ru. LOW leaves the chamber at ambiet temperature (i.e., chamber heater is off). MEDIUM heats the chamber to 3 C. HIGH heats the chamber to 65 C. 3. SPEEDVAC SC110 OPERATION 1. Load samples symmetrically ad verify that the rotor is firmly seated ad that the hold-dow kob is had-tight. 2. Close the cover ad set the CONCENTRATOR switch to ON. 3. Allow rotor to reach full speed (approximately 20 secods) before maually applyig vacuum with the bleeder valve. (Located o the top right pael,towards the rear of the istrumet). This valve exposes spiig samples i the sample chamber to vacuum from the vacuum system ad, at the ed of the ru, bleeds the chamber back to atmospheric pressure. Premature applicatio of vacuum ca cause bumpig ad foamig; i some cases, sample could boil out of the tubes.. Tur arrowhead idicator of bleeder valve toward cocetrator (OPEN) to apply vacuum. 5. Select DRYING RATE as described i sectio Cotiue the ru util a predetermied time has elapsed, or util a vacuum gauge, cofigured i-lie betwee the SpeedVac ad vapor trap,idicates a predetermied vapor pressure i the sample chamber. To devise a stadardized ed poit (time or vapor pressure) for a specific applicatio whe repeatability of results is importat, coduct oe or more test rus with expedable batches of sample. 7. At the ed of the ru, tur arrowhead idicator o bleeder valve toward the left (CLOSED) to bleed the sample chamber to atmosphere; allow 10 secods for pressurizatio.. Stop the ru by settig the CONCENTRATOR switch to the OFF positio. The rotor brakes util rotatio decreases to 6 RPM. Do ot ope the cover util rotatio has completely stopped. (All 20 V uits have a cover lock that prevets cover release at rotor speeds >6 RPM). 9. To shut off chamber heater, set DRYING RATE switch to LOW. 6

8 3.5 SPEEDVAC PLUS SC110A/SC210A OPERATION 1. Load samples symmetrically ad verify that the rotor is firmly seated ad the hold-dow kob is had-tight..0 SPECIFICATIONS 2. Close the cover ad set the CONCENTRATOR switch to ON. (Bleeder valve operatio i SpeedVac Plus models is automatic. A speed sesor iside the cocetrator applies vacuum whe the rotor reaches 00 RPM). 3. Select DRYING RATE as described i sectio Whe the ru is stopped by settig the CONCENTRATOR switch to the OFF positio, the uit immediately bleeds the rotor chamber, the waits 10 secods before brakig the rotor. Do ot ope the cover util rotatio has completely stopped. 5. Shut off chamber heater by settig DRYING RATE switch to LOW. 11 APPENDIX 1 MAINTENANCE AND SERVICE Maiteace: Maitaiig a clea istrumet is crucial for depedable results. Clea spills immediately. Dried solvets ca build up, impairig rotor rotatio. Periodic cleaig of the sample chamber prevets problems. Use a deterget solutio o a spoge or gauze, the thoroughly wipe the chamber dry. Excess moisture i the chamber is removed o the ext SpeedVac ru, but ca alter the performace of this ru. Keep the drive shaft ad rotor ceter lightly coated with vacuum grease. Do ot lubricate the cover O-rig. Upper Maget Assembly: Although the Upper Maget Assembly (UMA) is equipped with sealed bearigs, usig the SpeedVac with aggressive acids ad bases ca cause bearigs to corrode over time. Should this occur, the UMA may eed replacemet (frequecy depeds o idividual usage patters). Call Thermo Savat s Service Departmet ( , , or (Fax) ) for parts or repair. For the SC110/SC110A, order Savat part # UMA100. For the SC210A, order part # UMA200. To determie if bearigs are wor: 1. Ope the sample chamber cover ad uplug the istrumet. 2. Remove the rotor hold-dow kob, rotor, ad three screws (Phillips) o the chamber bottom moutig plate. This plate of blue-white plastic forms the top portio of the UMA, which houses the bearigs. 3. Replace the hold-dow kob ad usig the kob as a hadle, pull the drive shaft upward util the UMA comes free.. Pull carefully to release the magetic attractio betwee the UMA ad the Lower Maget Assembly (ot visible), located beeath the metal plate i the SpeedVac floor. 5. Hold the plastic moutig plate rigid ad attempt to pivot the maget (bottom portio of the UMA) aroud it. If there is excessive play (wobble) betwee the plate ad maget, replace with ew UMA. To replace the UMA: 1. Alig holes, the secure ew UMA with the three screws. 2. Replace rotor ad verify that it is seated properly o the aligig pis ad that the hold-dow kob is had-tight. 3. Recoect the istrumet to a appropriate electrical outlet ad resume operatio. 10

12 APPENDIX 2 APPLICATION INFORMATION Dislocatio due to brakig: Whe highly discrete solutes are dried usig ordiary rotors ad the total rotor mass is small, some users ote that the miute solute pellet is dislocated by the cocetrator s brakig actio, impedig recovery. For these critical applicatios, Thermo Savat recommeds the use of a heavier rotor. The coical tip of the sample tube swigs out horizotally as rotatio icreases. This draws solute to the tip of the tube, miimizig pellet dislocatio o brakig. Typical results: The followig data were gathered usig Thermo Savat s SC110 SpeedVac cocetrator, RVT100 (-55 C) Refrigerated Vapor Trap, VP100 (113 liter/mi) vacuum pump, the RH20- rotor, ad 20 glass tubes of x 75 mm, each cotaiig 2 ml of solvet. Time is show as hours:miutes. SOLVENT WATER METHANOL SETTING OF DRYING RATE SWITCH LOW MEDIUM HIGH 3:5 0:55 3:07 0:3 2:01 0:33 These approximate dryig times are preseted as a guide oly. Results will differ depedig o type of vacuum pump, temperature ad capacity of vapor trap, itegrity of seal betwee fittigs ad coectios, degree of cotamiatio, umber of tubes per ru, ad other factors. Further improvemets i evaporatio rate: Rotors with alumium tube carriers, such as the RH20-, coduct more heat to the samples ad ehace evaporatio. To speed evaporatio further, use a Thermo Savat Radiat Cover: For the SC110/SC110A, order the Thermo Savat RC110B Radiat Cover. For the SC210A, order the Thermo Savat RC210B Radiat Cover. WARNING: Never shie a lamp or heat lamp through the stadard acrylic chamber cover. The cover absorbs heat ad will deform, especially while the sample chamber is uder vacuum. Recofiguratio: The polypropylee vacuum fittigs do ot withstad strog lateral force. Should you ever eed to remove a vacuum lie, carefully pull straight out to slide the tubig off the fittig. DO NOT twist the tubig to remove it as this may damage or loose the fittig causig a vacuum leak. IF the tubig does ot come off whe pulled, CAREFULLY slit the existig tubig at the fittig (avoid scorig the fittig). Remove the tube ad discard it. Recofigure the SpeedVac ad system compoets as desired. To reattach the lie, place a thi film of vacuum grease (or use some soapy water) o the fittig; carefully slide ew tubig straight o over fittig. 11

13 APPENDIX 3 TIPS TO ENHANCE PERFORMANCE 1. TO INCREASE THE RATE OF SOLVENT EVAPORATION AND SHORTEN DRYING TIME, TRY ONE OR MORE OF THE FOLLOWING: Use glass tubes ad vials i place of plastic. Istall ad use a Radiat Cover o the SpeedVac. Preheat the SpeedVac chamber ad rotor usig MEDIUM DRYING RATE. Use rotors with metal tube carriers istead of plastic rotors (e.g., RH-20, RH-1). These rotors coduct ad trasfer heat to tubes for faster sample dryig. Prewash plastic tubes with methaol to remove materials that ihibit or prevet evaporatio of aqueous samples. 2. TO KEEP VACUUM PUMP OIL CLEAN, TRY THE FOLLOWING: Empty solvet(s) collected i the Refrigerated Vapor Trap as frequetly as possible. Chage the oil i the vacuum pump whe it becomes brow i color or smells of solvets beig evaporated. Attach a Thermo Savat VPOF110 Vacuum Pump Oil Filter/Recirculator to your vacuum pump whe evaporatig acids (e.g., HCl, acetic, TFA) i the SpeedVac. Attach a Thermo Savat SCT0 Chemical Trap with acid eutralizatio cartridge whe evaporatig acids (i.e. HCl, acetic, TFA) i the SpeedVac. If you are usig a gel dryer with your SpeedVac, remove the gel dryer from the system. Use a oil-free/maiteace-free Thermo Savat GelPump GEP10 for the gel dryer.
