6 Do not handle the power cord harshly. Disconnection or short may cause if you bend or put a heavy thing on the device. When you take a plug out in a wall socket, turn the power off and then take the plug out carefully. Insert the plug securely into a wall socket. Loose connections result in excessive heat generation to the device and may cause arcing to occur at the connection. Avoid using multi-point extension power cords. It may cause excessive heat generation and be the cause of a fire. If you extend the power cord, use the cord to have sufficient electric capacity. Be sure to operate the device only with the specified power supply. When you connect the product specified by 100V to 200V, it may be caused to over-heat, electric shock or a short circuit to each part. Do not connect wrong electricity since it is the cause of a fire. Disconnect the power supply plug if the unit is not going to be used for an extended period of time. 13 Installation 6 1. After receiving the unit and putting it on the specified place of use remove the wrapping. 2. After positioning the unit, lock the moving wheel 10 in order to make position fixed and the unit stable. 3. Check the unit whether there are eventual transport damages or not. In case there are some transport damages found please inform the forwarder and your contractor. If there is no damage, please go to next point Let the unit stand for a few minutes so that the all the internal parts and components of the unit can be stabilized. 5. Open the door at the left side of the unit. You can find a room for pipe assembly and electric distribution plane. Please check carefully if there is anything wrong including disconnection of the electric wire, breakage of pipe or fitting element. In case there is any kind of damage found, please inform the forwarder and your contractor. If you cannot find anything wrong, please go to next point For normal use of the unit, confirm that the valve 16 is open, and water drain valve closed. 7. Connect the main power cord to the socket. 8. (NOTE) If you bye WAC-80 or WAC-100, power consumption of the heater in the vessel is too high to use normal wall socket. (Typical wall socket in EU and Asian countries permits around 3kW maximum per socket) So, a big 3-pole plug (for single-phase 230V AC with ground) 13 comes with power cord, and a socket with wiring connector 14 is supplied together as seen in the picture on the page 10. You have to wire the power supply line of enough electric power (4kW for WAC-80 and 5kW for WAC-100 at least) to the supplied connector 14 before plugging. 9. 14 WARNING Place the device on a level, horizontal surface (floor) to support the capacity of the device without vibration. Do not install at a dangerous place. Do not operate the device where there are flammables. It could be caused serious accidents like explosion or fire. Do not operate the device outside. Avoid areas where the sun shines or rain directly on the device. It is very dangerous as cause over-heat or a short circuit. Do not tilt or lay down the device, and do not impact the body of the device. The device includes shock-sensitive pipes and fittings, which could be damaged by the tilt or impact. Do not install the device where it can get damp or dust. It may be caused overheating, a short circuit or a fire by damp or dust inside of the device. Do not use this device when temperature falls below 20 C and rise above 40 C. Do not use the device at a place with high temperature. Do not operate the device near a heater, blowing an air-conditioner. It may be caused an accident by overheating. The device should be set at a well ventilated place. Be sure the holes on the side or rear surface of the body are not blocked by a wall or an object. Do not operate the device at a poor ventilated place or use closed. It may cause a fire or damage of the device as not to release heat or insufficient ventilation may result. Set the device a lightening place. 15 Cautions in Use 7 WARNING Be careful to operate the device. You MUST read this manual carefully and completely before using. The device should only be operated by laboratory personnel especially trained for this purpose and familiar with all precautionary measures required for working in a laboratory. If unexpected sound, smell or smog is generated after connecting electric power, pull out the main plug and contact manufacturer or a supplier. Do not put any objects insides. When an object that is easy to be combustible is like a stone, a metal or paper goes insides, it may cause a fire, an electric shock or a break down. If do, pull out the mains plug from a socket and contact a manufacturer or a supplier. Do not disassemble or remodeling by yourself. It may cause damage to the designed safety. We do not have any duties for an accident, quality deterioration or a defect caused by a customer. All the mechanisms and electric/electronic devices in this machine are designed and developed by the manufacturer. The devices could be disassembled and serviced only by the engineers of manufacturer or by engineers approved by manufacturer. Do not touch the device with wet hand while operation. Do not turn on and off frequently. It could damage the compressor by overload. This device must be connected to an earthed power supply. 16 CAUTION Only demineralised or distilled water MUST be used. If dirty water is used, it will cause damage of all the components inside the vessel, inner surface of the vessel, pipes, fittings and valves. Do not use tap water! Prior to sterilization, check sterilizing chamber to ensure there is sufficient feed water. No sterilization without feed water! WiseClave WAC Autoclaves have been designed for the following purposes; For sterilizing glassware, rubber items, pipette tips, instruments, filter bodies, as well as consumables, waste in bags, and to destroy infectious or septic materials. For sterilizing liquids, solutions and culture mediums in open or lightly closed vessels. For sterilizing foodstuffs For streaming liquids or for dissolving agar. For non-pressurized disinfection of heat-sensitive culture media and solutions. Liquids and foods must not be sterilized in pressure-sealed containers. Easily combustible or toxic liquids must no be sterilized The following products are not suitable for sterilization; Wooden items Bonded parts Plastic materials sensitive to hot water (e.g., screw tops) Electrical equipment Vessels or instruments made from steel or iron Metering equipment, unless their suitability for stream sterilization is specifically mentioned Destruction bags must be open during sterilization, otherwise they may burst. 17 Before every sterilizing operation, check that the lid stiffener 2 on the lid 4 has moved all the way into the lid clamp 3. And check the lid screw handle 1 is turned until it cannot be turned anymore. Do not open sterilizing chamber until full sterilization operation cycle is finished. Allow chamber pressure gauge to slow zero before opening the autoclave. Watch for hot steam and hot water! Hot steam may escape when the lid is opened. The lid handle should only be gripped at the experiment end. Step back when opening the sterilizer. You must wear the following protective items when opening the sterilizing chamber and removing the sterilized articles; Apron Eye protectors Heat insulated gloves Watch for hot surfaces and hot steam! OPT sensor (SAFETY) knob should be set as operating temperature +20 ~ +30 C. Please be noted that the heater doesn t work if the knob is set to 0 position. When sterilizing liquids, do not fill vessels more than two-thirds full, as otherwise liquids may boil over. Do not sterilize pressure-sealed vessels. Explosion hazard! Watch for broken glasses! 18 After sterilizing liquids; Danger from retardation of boiling. For open vessels containing up to 1 liter: wait at least 2.5 hours For open vessels containing up to 2 liters: wait at least 3 hours Do not remove flasks or bags until the chamber temperature drops below 70 C. Do not hang onto or lean against the open lid. DO not climb onto the autoclave, as this may cause the autoclave to tilt. Please avoid ultrasonic lamps. The unit could be cracked or faded. Disconnect the power supply plug if the unit is not going to be used for an extended period of time. This Symbol indicates that this product is to be collected separately. 20 Operation 9 1. Preparation for Use DAIHAN Autoclave is a kind of Steam Sterilizer. So the user should fill water into the pressure vessel before operating the unit. (NOTE) Distilled water should be used. The user should pour water to the pressure vessel, so the bottom part of the vessel (bottom cover) should sink under the water. (NOTE) If the water level is too low, the machine cannot start sterilization operation by the functioning of water level sensor equipped in the vessel. (NOTE) Closing of manual ventilation valve 7 must be checked. (NOTE) Opening of valve 16 must be checked as well. This valve can be closed only for the diagnostic purpose by an authorized engineer. Connect the power cord to the proper power socket. (NOTE) If you bye WAC-80 or WAC-100, power consumption of the heater in the vessel is too high to use normal wall socket. So, a big 3-pole plug (for single-phase 230V AC with ground) 13 comes with power cord, and a socket with wiring connector 14 is supplied together as seen in the picture. You have to wire the power line of enough electric power to the connector 14 before plugging in order to use the machine. Turn on the Main Switch 11. At this moment, if the water level in the vessel is not sufficient, LCD shows Low-Level error message with buzzer sound. In this case, supply more water until you can see the normal status (Set Timer mode) on the LCD. (NOTE) Set the OPT sensor knob 15 to 20~30 C higher than operation temperature (eg., If your operating temperature would be 121 C, proper value of the OPT sensor would be 150 C). Please be noted that the temperature on the scale is rough values. 2. Preparation and Placing of Materials All materials autoclaved must be placed in an autoclavable tray (wire basket delivered together with machine) with sides that are at least 5 inches in height. The use of trays should eliminate anything boiling over and spilling into the autoclave. Therefore, all materials must fit within this tray. Only autoclavable materials may be placed in the autoclave. If you are unsure if something is autoclavable, check with the manufacturer. Liquids to be autoclaved must be in an autoclavable vessel that is at least twice as large as the volume to be autoclaved (i.e. If you are autoclaving 1 liter of media, you need to put it in a flask that hold at least 2 liters). 21 must be slightly loose so that pressure created during the cycle does not cause the vessel to break. For screw cap containers you can make the lid hand tight and then loosen the lid by one-half turn. Dry goods (solids) must be placed in an autoclavable tray with 5-inch sides also. Garbage must be put in autoclavable bags and inside the autoclavable trays with 5-inch sides. Sharps containers must also go into the same type of autoclavable trays. To ensure proper sterilization of both garbage and sharps containers, 1 liter of water should be added to the bags or sharps containers. This water will vaporize during the autoclave cycle to allow better penetration of steam and more efficient sterilization. Autoclave garbage bags must be tied off. (WARNING) Use of non-autoclavable trays and containers may result in injury to the user and damage to the equipment. After placing the material to be autoclaved, close the lid 4 of pressure vessel by turning the lid handle 1 clockwise until it cannot be turned anymore. 3. 22 4. Set Operation Cycle Mode WiseClave WAC autoclaves offer two different operation cycles named SOLID cycle mode and LIQUID cycle mode. As the name implies, LIQUID cycle is for autoclaving liquids while the SOLID cycle is for autoclaving solid or dry materials. With SOLID cycle mode operation, steam is ventilated out quickly after the completion of sterilization. On the other hand, with LIQUID cycle mode, steam is ventilated out very slowly after sterilization. The following table shows you the difference of the two cycles and proper use; Type of Cycle SOLID or fast exhaust LIQUID of slow exhaust Appropriate Material Dry goods, glassware, etc. Liquids Description This cycle charges the chamber with steam and holds it at a set pressure and temperature for a set period of time. At the end of the cycle, a valve opens and the chamber rapidly returns to atmospheric pressure. This cycle prevents sterilized liquids from boiling. Steam is exhausted slowly at the end of the cycle, allowing the liquids (which will be super-heated) to cool. By pushing MODE button 19 and SUB button 20 sequentially, you can enter into the Set Operation Cycle mode. Factory default mode is SOLID cycle. By pushing jog-shuttle knob 24, you can change the cycle, which is toggled by pushing repeatedly. Cycle shown on the LCD currently is the selected operation cycle. 5. Setting Timer 5.1 Set Operation Timer The initial mode after power-on is Set Operation Timer mode. TIMER lamp 21 is blinking. (NOTE) Operation Time means the Sterilization time. This timer begins to run (count down) when the Temp PV (present temperature) reaches the Temp SV (set temperature). Tim SV on LCD indicates set value of operation timer and Tim PV indicates present value (remaining time) of the timer. (NOTE) Initial value (SV) of the timer is 00:00, which means infinite time. With zero value of Timer SV, the machine cannot start operation. Turn the Jog-shuttle knob 24 then SV and PV change together. SV can be set in 1 minute, and the maximum value is 99:59 (99 hours and 59 minutes). Set required time by turning the Jog Knob 24 and PUSH it to complete the timer setting. 23 (NOTE) By pushing the jog-knob 24 with set value, the timer begins to operate but doesn t start actual counting before beginning sterilization. It counts down when the Temp PV (present temperature) reaches the Temp SV (set temperature) as mentioned above. The machine stops operation (heating) after the SV of timer has passed. 5.2 Set Delay Timer It turns to Set Delay Timer mode by pushing SUB button 20 in Set Operation Timer mode. TIMER lamp 21 is blinking. DTim SV on LCD indicates set value of delay timer and DTim PV indicates present value (remaining time) of the timer. Set required time using the Jog Knob 24 and push it to complete the timer setting. Then timer is started and TIMER lamp 21 is on. The machine starts operation (heating) after the SV of delay timer has passed. It returns to Set Operation Timer mode by pushing MAIN button (NOTE) While the machine is being operated already (by setting operation timer, temperature and heating start), delay timer operation cannot be set and started Set Temperature 6.1 Set Temperature It turns to Set Temperature mode by pushing MAIN button 19 in Set Operation Timer mode. HEAT lamp 22 is blinking. Temp SV on LCD indicates set value of operation temperature and Temp PV indicates present value (measured value by temperature sensor in the pressure vessel) of the temperature. Turn the Jog-shuttle knob 24 then SV changes. SV can be set with the resolution of 0.1 C. The maximum value is C and the minimum value is 25.0 C. Set required operating temperature by turning the Jog Knob 7 and PUSH it to complete the temperature setting. The controller starts operation of heater in order to reach the set temperature. HEAT lamp 22 is on. (NOTE) As mentioned in the section 5.1, the machine cannot start heating (sterilization) operation without setting the operation timer to non-zero value. 6.2 Temperature Offset WiseClave WAC autoclaves intelligently and precisely controls the temperature inside the pressure vessel by digital Fuzzy control system, so high precision with low over/undershoot is assured. But, some users want to synchronize the temperature PV of the machine to the temperature value measured by other thermometer that is used as a reference meter for the DAIHAN Scientific, WiseClave WAC Steam Sterilizers Digital Fuzzy-controlled Autoclaves. 23/51

24 whole experimental process. In such a case, WAC autoclaves offer a function of adjusting the temperature PV within 10.0 ~ C User s self-compensation function. It turns to Set Offset Value mode by pushing SUB button 20 in Set Temperature mode. Temp PV on LCD indicates current temperature and Offset indicates the value to be added to PV temperature (compensation value). Turn the Jog-shuttle knob 24 then Offset and Temp PV changes together. Offset can be set with the resolution of 0.1 C, and input range is 10.0 to C. Temp PV shows value added or subtracted by Offset value. It returns to Set Temperature mode by pushing MAIN button Locking Mode It turns to Locking mode by pushing MAIN button 19 in Set Temperature mode. In this mode, turning or pushing Jog knob doesn t work. So, you could prevent any changes of set values by mistake. Operation Timer PV and Temp PV are shown on LCD. It returns to Set Operation Timer mode by pushing MAIN button End of Sterilization When the set time has passed and the sterilization is finished, firstly heating is stopped, and then, the steam and pressure is exhausted out and the pressure is removed. User can identify this by buzzer sound for 30 seconds. As described in the section 4 of this chapter, the way of exhausting steam and pressure is different based on the operation cycle. In SOLID cycle mode, valve is opened completely and steam is discharged quickly. In LIQUID cycle mode, valve is not completely opened at a time. Firstly it waits for 5 minutes with valve closed after heater OFF. And then, the valve is opened during 3 seconds, after that the valve is closed again for 2 minutes, and then it reopens during 3 seconds, etc. This procedure repeats until the temperature comes down to 90 C (lasts approx. 30minutes), and then it waits until the temperature reaches down to 80 C with valve closed again. After that, the valve is closed completely. While steam is discharged out of the pressure vessel, the user cannot see the steam out. Steam in the vessel is converted into water by passing trough a specially designed steam condenser. This feature offers safety and convenience to the user. 25 9. After Sterilization When the sterilization is completed, pressure is exhausted through steam discharging automatically. You could check it with the pressure gauge 5 next to control panel. After the pressure comes down to 0 ~ 0.2 kg/cm 2, you may open the lid by turning lid handle counterclockwise. If you want to confirm the complete ventilation of the steam and pressure, you may use the manual ventilation valve 7 at this moment. If you want to remove the water remained in the vessel, you could do it with the drain valve. User can open the door at the left side of the body, and can find the water drain valve 17 there. Connect a kind of tube to the valve and open it, then the water remained in the vessel can be poured out. 10. Storage Function In each setting mode (Set Operation Timer mode, Set Delay Timer mode, Set Temperature mode, Set Offset Value mode and Set Operation Cycle mode), you can store the current set values by simply pushing the Jog knob for 1 second. For showing the value is successfully stored, the set value on LCD blinks three times. 30 Troubles and Solutions 13 Situation Confirmations and Solutions Power failure Check the power cord is plugged. Check the supplied electricity is proper and regulated. Check the Leakage breaker is ON. Not heating Check the temp SV and confirm the operation started by pushing Jog knob. Check the MAIN power switch is ON. Check the SAFETY knob is set to too lower value than recommended value and not set to 0. Heating but it doesn t reach the SV temperature Problem in the heater or temperature sensor. Please call technical service. Noise The noise could be generated by the fan equipped in the steam condenser. If it becomes too big, please call technical service.
