2 GENERAL OVERVIEW Thank you for choosing BioReactor Sciences (BRS). Mechanically Agitated (MA) bioreactor systems are the most common and universal type of bioreactors available today. Our MA bioreactor is distinguished from other systems in terms of construction and design. Our modular design, open sourced software, and use of industry-standard materials & components result in durability, simple installation, straightforward use, and uncomplicated maintenance. This quick reference guide contains the necessary information to get started using the BRS MA bioreactor system. Please retain it as a reference guide to ensure proper use. If needed, additional copies can be obtained on our website. DEVICE OVERVIEW PRINCIPLES OF OPERATION Switching the Control Console on/off On: the panel may be switched on by turning on the power to the entire MA system via the red power switch in the back of the system. Off: the panel may be switched off by turning the power off to the entire MA system via the red power switch in the back of the system. HMI / User Interface Users can use the HMI display panel to implement the following functions: Creation, editing, and adapting of cell culture programs Execution of cell culture programs Manual control Read-in and read-out of programs and data Display and troubleshoot alarms The user interface is a graphical overview of the reactor, with symbols for pumps, and if applicable 2
3 additional peripherals with their typical locations relative to the reactor. The user interface is divided into 2 general sections: the work area and the sidebar. Below is the Main screen illustrating the work area, the sidebar, and general symbols. The sidebar will always be present showing the date, time, and if applicable, the loaded program. Date, Time Example: 18/02/15; 11:31:49 Date in Format: [dd/mm/yyyy]; Time in Format [hh:mm:ss] 24 hour clock / military time Screen Title ( Main ) Date and Time Program name or number Login/Logout info for different user access Sidebar Work Area Overview of the Main Function Buttons and Screens Button / Screen Description Start screen with graphical overview of the controlled bioreactor: Display of components with current status (ON or OFF) Overview of process parameters and their current setting and output/reading Access to important menus for operating input Displays Pump & Gas settings. Respective Pump & Gas accessed by selection of either Pump or Gas. 3
4 Access activation and manual/override controls for system components. Select from: Current Program current program running/loaded On-File Program saved programs on-file for access Feed Program Current current feed program running/loaded Feed Program On-File - saved feed programs on-file for access Display of process sequences, selection of 6 different components: agitator (AG), AIR, ph, DO, temperature, feed load cell. Note: to access individual component trend, click on individual component on Main screen. Display of system operations with date, time, and message. Scroll up and down to view past operations. Clear and save-todisk operation messages as needed. Access to System Tools and System Configuration. System Tools access to Login, Log out, Brightness, Panel Setup and other system tools System Configuration access various system parameters for agitator, ph, AIR, Pressure, etc. control Current, Alarm Control, and Alarm Records Current Alarm displays current and recent alarms. Alarm Control Set Hi, Lo, and Time (secs) for various alarms Alarm Records Store and clear alarm records 4
5 Screen Descriptions Below is a flow diagram outlining the HMI process structure. Main Screen Click on any system component (Agit, Foam, Feed, AIR, Temp, DO, or ph) to access its Calibration, Control (PID), Settings, etc. screen 5
6 Clicking on the ph component produces a drop down menu to access ph Calibration and Control (PID) Click on Main to produce a drop down menu to access the 8 primary screens Manufacturer Operating Environment Requirements: Operating Temperature: 0 C 60 C Storage Temperature: 20 C 70 C Relative Humidity: 5% 95% non condensing 6
7 TREND MENU To collect data, you must first ensure that the system is recording said data. To accomplish this: 1. Activate the TREND function by pressing the TREND button on the CTRL PNL. This will initiate the recording and collection of data. 2. User must then set the maximum and minimum ranges for each parameter. To do so, go to the TREND screen (press TREND button on MAIN side bar) and enter the y-max and y-min for each parameter. Each parameter is designated by a color per the following: Green = AG (agitator) Yellow = TP (temperature) Pink = ph Brown = DO (dissolved oxygen) Blue = AIR Purple = FD (feed) 3. Then enter in x-scale or Time to adjust the x-axis accordingly. 4. The data will then continuously log and can be viewed either in graphical form on the TREND screen or in data points form on the DATA screen. 5. To save the data, press the SAVE button on the DATA screen. Data will transfer in Microsoft Excel form. Be sure to not turn the system off until the data is completely saved on your computer or USB flash drive. MAINTENANCE & CLEANING BRS Pinch Valves are designed for normal spray down sterilization procedures and include internal splash seals that prevent fluid spray penetration. Normal biological sterilization procedures can be done to the exposed valve head surface areas that come in contact with the tubing (i.e. tube slot area). Some common sterilization solutions or methods that can be used at appropriate concentrations are: Ethanol (3 5%) Methanol (3 5%) Iso Propyl Alcohol (3 5%) Hydrogen Peroxide (3 5%) Ammonia (3 5%) Bleach UV radiation exposure not exceeding 15 minutes ALARMS The MA-100 will alarm (indicated by the lower right hand Alarm button flashing) when any errors or faults occur. Error or faults may include any parameters exceeding their high/low set points or mechanical faults. Pressing the flashing Alarm button will take user to the Alarm_Current screen to learn more about the error or fault. 7
8 The Alarm_Current screen below allows the user to turn alarms on and off, to read the alarm history, and to acknowledge any alarms while it is active. Alarm_Current Screen The Alarm_Current, Alarm_CTRL, and Alarm_Record screens can all be accessed from the Main screen by pressing the Alarm button that will produce the Alarm Sub menu, shown below Alarm Sub Menu The Alarm_CTRL screen below allows user to set the high threshold (HI), low threshold (LO) and the duration of time the parameter must exceed the threshold before the alarm triggers (Time [sec]) settings. HI: setting + HI setting is upper limit threshold/range of alarm LO: settings LO setting is low limit threshold/range of alarm. Time (sec): duration of time in seconds, the parameter must exceed the threshold setting before the alarm triggers. 8
9 Alarm_CTRL Screen The Alarm_Record screen below allows the user to access the log of alarms cataloged by Date and Time. Users may scroll up or down to find the desired alarm and record the Status and Message of the respective alarm. User may clear the alarm record by pressing the Clear button, and save the record by pressing the Save button (record will save to device user has connected [USB flash drive, PC, etc.].
