Short Term Scientific Missions (STSM) Report COST action FA1104: Sustainable production of high-quality cherries for the European market COST STSM Reference Number: COST-STSM-FA STMS Title: Planting, Pruning and Hybridization of Cherry Periods: 06 April April 2015 Visitor Adem DOGAN Akdeniz University Faculty of Agriculture Department of Horticulture Antalya Turkey Mobile Phone Host: Prof. Dr. Vladislav Ognjanov Novi Sad University Faculty of Agriculture Novi Sad, Serbia Phone:

Purpose of STSM The purpose of this STSM was to improve collaboration and exchange information about the cherry culture: breeding, planting and pruning technology, selection of dwarfing rootstocks for sweet and sour cherry, development of intensive cultivation technology on the principles of sustainable agriculture, aplication of in vitro and PCR techniques. Description of the work carried out during the short-term scientific mission In the first day of Novi Sad University, we met with Prof. Dr. Vladislav Ognjanov and his assistants Goran Barać, Jovana Dulić and Maja Miodragović. They introduced me to Faculty of Agriculture, Department for fruit growing and laboratories. We made detailed plan for my STSM. 1- Tissue culture Hybridization of Prunus avium L. is considered as a major path for development of new cultivars. Breeding program of Prunus avium L. is usually accompanied by the problem of low seed germination that is very difficult to achieve using conventional methods, especially in early ripening varieties. They showed me their embryo culture and developed techniques. At the same time, anther cultures, variety Regina, was ready for first subculture. Firstly we prepared medium of liquid culture medium supplemented with (mg liter 1) 100 myoinositol, 1 thiamine-hcl, 1 nicotinic acid, 1 pyridoxine HCl, and with 3% sucrose, 4.4 µm BA, and 4.5 µm 2,4-D; ph = 5.0. And after the sterilization, we prepared the petri dishes with 9 ml media.

Haploid callus was separated from anthers and transferred to fresh medium using microscope. In vitro culture of sour cherry embryos: they show me how to handle explant and transferring to new medium. 2- Hybridization Anthers of male parents Rita and Early Star sweet cherry were collected from main growing cherry region in Serbia, Belgrade region, village Brestovik. Female parents were Bigarreau hatif burlat, Carmen, Valery Chkalov - mother trees were located at scientific field, Rimski Šančevi and Kać After flowers were collected at balloon stage, the pollen was handled. Anthers were separated from flowers with combs and dried. In the field, numbers of flowers were calculated on each isolated branch. Pollination was done with hand and flowers immediately closed with paper bag.

After the 10 days later, we checked the situation of fruit set. 3- PCR Technics They also showed me to their PCR technics. The PCR was used for biodiversity analysis of cherry accessions using SSR markers. 4- Planting We also organized last year s hybrids for planting in the orchard.

Regularly I had a lectures with professor Ognjanov. He presented their power point presentations; 1- Effect of pruning system, planting density and rootstock on intensive cherry production, 2- Progress in Cherry Rootstock and Cultivar Research, 3- General information about the breeding program in apple, pear, peach and plum. I presented Horticultural overview of Turkey and our project The Effects of Ozone and Different Packaging Systems on Postharvest Quality of 0900 Ziraat Cherries for Increasing Cherry Export. Prof. Dr. Vladislav Ognjanov organized four training trips for me; 1- He presented their planting, pruning and breeding programs. After presentations, we visited Research area of Novi Sad University (Rımskı Sancevı) and we saw collections of planting rootstock, sweet cherry, sour cherry, apple, apricot, peach. Many practical information in the orchards were given. 2- We visited Feketic which is very important place for sour cherry in terms of genetically diversity. At the same day, we met with Dr. Janos Apostol from Hungary. We talked about cherry selection factor and also we visited collections of sour cherry. On our way back we visited one commercial cherry producer.

3- They organized one trip to Fruska Gora. We visited commercial storage facility of ATOS Fructum and their apple orchards. We visited viticulture research area of Faculty of Agriculture, University of Novi Sad. Acknowledgments I wish to acknowledge the financial support given by the COST. And I thank for help and advice chair of action Dr. José Quero Garcia, STSM coordinator Prof. Dr. Francesco Paolo Marra and the host Prof. Dr. Vladislav Ognjanov and his assistants.
