The PX1 PCR Plate Sealer uses heat ad pressure to seal the plate ad sealig film together. Some sample materials may be iappropriate for a heat sealig process. For example, materials that are highly volatile or have low flash poits may be at risk of explodig or igitig durig the sealig process. WARNING! Risk of splashig. Always wear appropriate protectio agaist splashig. The PX1 PCR Plate Sealer uses heat to fuse the plate ad sealig film together. Expasio of air ad materials sealed i the sample plate could cause the seal or sealig film to rupture, dispersig the sample material i ad aroud the istrumet. This is especially true if the plate is jammed, stuck, or left i the sealig bay for a exteded time. WARNING! The plate, sealig film, plate support block, ad sealig frame may be hot after a sealig cycle or if left i the istrumet for a exteded period of time. Allow the plate ad accessories to cool before hadlig. WARNING! The display pael is made of glass ad cotais a liquid crystal substace. If the display is damaged, the glass ca break ito shards ad the liquid crystal substace may leak out. The glass shards are sharp ad ca cut ski or other soft materials. The liquid crystal substace is toxic. Do ot allow glass shards or liquid crystal substace to get i your mouth or eyes. 8 1 Itroductio Chapter 1 Itroductio The Bio-Rad PX1 PCR plate sealer is desiged to deliver cosistet ad reliable microplate sealig that withstads the stresses of thermal cyclig. The PX1 virtually elimiates sample evaporatio from the PCR experimetal workflow. 9 Chapter 1 Itroductio Plate (Figure 3) the metal plate is precisely heated to the programmed temperature. Durig sealig, the plate is lowered to efficietly seal a plate usig heat ad pressure. Remove the plate support block ad sealig frame whe the istrumet is tured o (Figure 7). Touch the butto to ope the drawer ad esure that both the plate support block ad sealig frame are removed from the istrumet. Oce removed, touch OK. The istrumet will automatically begi heatig to the last set temperature. Fig. 7. Remove support block. WARNING! Bur Hazard! Do ot leave the plate support block or sealig frame i the heated istrumet. They may become hot ad pose a hazard to the user ad experimetal samples. 2. Modify seal temperature ad time O the home scree touch Set Temperature Eter desired seal temperature betwee 100 ad 190 o C. Touch OK The plate automatically begis to heat to the ew set temperature. If the set temperature is lower tha the curret plate temperature, the heater will remai off ad the plate will cool passively The Off butto o the umeric keypad will tur the heater off O the home scree touch Set Time Eter desired sealig time betwee 0.5 ad 10 secods. Touch OK Note: Suggested sealig parameters are provided i Chapter 5 ad i Bio-Rad heat seal packagig. Refer to the Optimizig Heat Sealig sectio for istructios o optimizig these parameters. 4

11 Chapter 2 Usig the PX1 PCR Plate Sealer 3. Seal the plate (Figure 8). A. Determie correct side of the reversible 96/384- well plate support block to use (A). 96-well plates, side with the 96 recesses up 384-well plates, flat side up WARNING! Usig the icorrect side of the plate support block may result i poor sealig ad may possibly damage the istrumet. B. Touch to ope the drawer. Place plate support block i the drawer (B). C. Place PCR plate i plate support block (C). D. Place seal o plate (D). The yellow sticker o the Bio-Rad heat seal bag idetifies the sealig surface. E. Some seals (icludig Permaet Clear Heat Seal ad Peelable Foil Heat Seal) require the sealig frame to hold the seal i place. If required, place sealig frame o top of seal, aligig frame sides with outer grooves o the drawer guides (E). F. Touch Seal whe the set temperature has bee reached ad the seal butto is gree. The drawer will close ad the plate will be sealed. Whe sealig is complete, the drawer will automatically ope ad the plate ca be removed. WARNING! The plate ad seal remai hot for a few secods after sealig. Use cautio whe removig the plates. Tip: Keep the plate support block ad sealig frame outside the istrumet betwee sealigs so they do ot become hot. Alteratively, the drawer ca be kept ope to prevet the block ad frame from heatig up. 12 Chapter 2 Usig the PX1 PCR Plate Sealer Optimizig Heat Sealig Sealig optimizatio is a crucial step to esure a high-quality seal. Sealig a plate for too log or at too high a temperature ca result i oversealig ad potetially affect the quality of data i optical assays, icludig qpcr. Udersealig a plate may permit sample evaporatio. Tip: Optimize sealig coditios usig a empty plate. 1. Seal a empty plate with the recommeded sealig parameters listed i Chapter Carefully examie the etire seal ad plate surface. Look for eve sealig aroud each idividual well ad across the etire plate (Figure 9). 3. Make sure well rims are ot sigificatly deformed. Note: Miimizig well deformatio is especially importat for qpcr assays. Sigificat well deformatio could affect optical readigs. 4. If usig a peelable seal, peel off the seal ad examie the adhesive patters where the wells cotacted the seal. Look for uiform circular imprits across the etire seal (Figure 10). 5. Optioal: fill a sample plate with a colored liquid such as 0.01% bromopheol blue solutio. Examie sample volumes to esure pipettig accuracy. Seal the plate. Thermal cycle the plate ad examie all wells for sigs of evaporatio (Figure 11). 6. Save optimized sealig protocol so that it ca be easily accessed for future experimets. 13 Chapter 2 Usig the PX1 PCR Plate Sealer Storig ad Usig Sealig Protocols Storig Protocols 1. Touch Protocols o the home scree (Figure 6). 2. A scree displayig a table of saved protocols is displayed (Figure 12). View additioal protocols by touchig the ad arrows. 3. Touch the protocol you would like to edit or replace with a ew sealig protocol. 4. Touch Edit. A scree displayig the protocol ame, temperature, ad time appears. 5. Touch the parameter you wish to modify ad use the keypad to make the desired chages. Touch OK to save the chages or Cacel to revert back to previously saved protocol. Usig saved protocols 1. Touch Protocols o the home scree. 2. A scree displayig a table of saved protocols is displayed. View additioal protocols by touchig the ad arrows. 3. Touch the protocol you would like to use. Set Auto Shutdow The Set Auto Shutdow fuctio allows you to customize the duratio of time a idle istrumet remais at the set temperature prior to automatically shuttig dow. 1. Touch Set Auto Shutdow o the home scree (Figure 6). 2. Eter the time the idle istrumet should remai at the set temperature before automatically shuttig dow. Times ca rage from 30 miutes to 3 hours. If the istrumet has received o user iput for this set amout of time, the heater ad scree will automatically shut off ad the LED light will chage from gree to orage Touch the scree to brig the istrumet out of Auto Shutdow mode. The istrumet will automatically begi heatig to the last set temperature Chages made to the Auto Shutdow settig are saved whe the mai power is shut off 7

14 3 PX1 PCR Plate Sealer Maiteace PX1 PCR Plate Sealer Maiteace Periodically remove ay debris, dirt, or residues that might iterfere with proper istrumet fuctio or preset a chemical, biological, or radiological hazard. For istructios o hadlig ad cleaig radioactive or biohazardous materials, cosult the guidelies for radiatio safety ad biosafety provided by your istitutio. Cleaig Outer Istrumet Parts Tur off ad uplug istrumet. Allow istrumet to cool for at least oe hour. Exteral Casig 1. Use a damp cloth or tissue to clea spills off the outside of the case. Use a 10% bleach solutio or 70% ethaol solutio for decotamiatio. 2. Allow casig to dry completely before rebootig the istrumet. Touch Scree 1. Getly clea the scree with ay approved LCD cleaig aget. 2. Allow scree to dry completely before rebootig the istrumet. Exteral vetilatio slots Remove dust ad debris with a soft brush, damp cloth, or vacuum cleaer. Cleaig the Plate The plate (Figure 3) was desiged with a ostick surface to facilitate cleaig. This feature makes it easy to remove seals accidetly placed upside dow durig sealig. 1. Tur off ad uplug the istrumet. 2. Wait at least oe hour for the istrumet to completely cool. Cautio: The plate may retai heat for more tha oe hour. Bio-Rad recommeds cleaig the plate whe the drawer remais o its tracks. If this is too difficult, see the Removig the Drawer sectio. 3. If removig a upside dow seal, getly pull the seal off the plate oce the plate has fully cooled. 4. Wipe the plate with a paper towel or lit-free wipe dampeed slightly with water. Ay adhesive or melted plastic o the plate should easily wipe away. Note: Never use abrasives or sharp-edged objects to clea the plate as they may damage the ostick surface. 5. Allow the istrumet to dry. 8

15 Chapter 3 PX1 PCR Plate Sealer Maiteace Cleaig the Drawer Use a damp cloth to clea up spills iside the drawer. Decotamiatio procedures should use a 70% ethaol or 10% bleach solutio. Use a silicoe-based grease to lubricate the rails o the bottom of the tray if ecessary after cleaig. Lithium- ad molybdeumbased greases should be avoided as they may react with the silicoe-based grease. Removig the Drawer 1. Tur off ad uplug the istrumet. 2. Allow the istrumet to cool for at least oe hour before removig the drawer. WARNING! Bur Hazard: The plate ca remai hot loger tha a hour ad ca bur the user if touched whe hot. Allow the istrumet to cool completely before performig ay maiteace. 3. Maually ope the drawer. 4. Reach uder the bottom of the ope drawer (Figure 14) ad push both tabs to the left. Figure 15 shows the uderside of the drawer. (Black tabs should be pushed to the left whe the istrumet faces up.) 5. While pushig both tabs to the left, pull the drawer off the tracks. The tracks will exted slightly as the drawer is pulled. Avoid touchig the rails because the grease may be removed. The rails ca be wiped with a soft dry cloth. Decotamiatio procedures should use a 70% ethaol or 10% bleach solutio. If a liquid decotamiatio aget is used, the tracks should be lubricated usig a silicoe-based grease. Lithium- ad molybdeum-based greases should be avoided as they may react with the silicoe-based grease. Replacig the Drawer 1. Make sure the drawer tracks are fully exteded (Figure 16). Fig. 14. Place metal progs o the rear of the drawer iside the black eds of the tracks (Figure 17). 3. Maually push the drawer i all the way to the closed positio. If the drawer appears crooked, esure rails are properly seated o the tracks before forcig the drawer i completely. Fig. 17. Replace the drawer. If ay issues are detected, the istrumet will display a error message. I some cases, the user will be istructed to perform a fuctio to clear the error. After performig this fuctio, touch OK. Although fuses should ever eed to be replaced, some istitutios prefer to replace fuses o a regular basis to maitai uiterrupted operatio. If the PX1 does ot tur o, first cofirm the power cord is i a fuctioig power source withi the istrumet specificatios. If the cord ad AC power source are fully fuctioal, use the directios below to determie if the fuses are itact. WARNING! To prevet electrical shock, always tur off ad uplug the istrumet before checkig the fuses. WARNING! The fuses ad other protectio features have bee desiged ot to fail over the life of the istrumet. If the fuses fail more tha oce, there may be a electrical fault i the istrumet. If this occurs, cotact Bio-Rad customer support for assistace. Directios to Replace a Fuse (Figure 18) 1. Tur off ad uplug the istrumet. Remove power cord from the istrumet. 2. Use a sturdy blut object to getly pry ope the fuse cover (A). 3. Use a small coi to useat the fuse carrier (B). 4. Remove both fuses from the fuse carrier ad ispect. If the wire is o loger itact, the fuse is blow (C). 5. If either fuse is blow, istall two ew fuses (8 A, 250 V, 5 x 20 mm, Fast Blow) ito the fuse carrier (see Table 2). 6. Reistall the fuse carrier usig firm pressure to esure it is fully seated. 7.
