1 Standard Operating Prcedure Centrifuge Safety SOP Number: SOP-CS-01 Categry: Lab Equipment Supersedes: N/A Effective Date: December 1, 2017 Pages 8 Subject: CENTRIFUGE SAFETY Prepared By: Lisa McAvy 16/Nv/17 dd/mmm/yy Apprved By: Vernica Harris-McAllister 17/Nv/17 dd/mmm/yy 1.0 POLICY This prcedure describes the standard peratin fr using the ambient and refrigerated centrifuges lcated in the Research Centrifuge Rm (Cnnell 4, Rm ) in the W.J. Hendersn Centre fr Patient-Oriented Research (WJHCPOR). The centrifuges are delicate pieces f equipment. Rtrs n centrifuge units are subject t mechanical stress that can result in rtr failure. In additin, imprper lading and balancing f the rtr can cause it t break lse while spinning. Fr these reasns, the centrifuge must be prperly used and maintained. Failure t fllw prper peratin f the centrifuges may nt nly result in damage t the equipment and bilgical samples, but als cause injury t the user wh is perating it. 2.0 PURPOSE All users f the centrifuges in the WJHCPOR are required t be trained by a designated Kingstn General Health Research Institute (KGHRI) staff member prir t using the centrifuges in rder t familiarize themselves with the functin, specificatins, peratin and rutine peratr care and maintenance f the centrifuges t ensure safety. 2.0 RESPONSIBILITY Users are respnsible fr: Attending rientatin and training n the ambient temperature and refrigerated temperature centrifuges. Training will include lab safety and use and care and maintenance f the centrifuges. Training is mandatry and is required t be cmpleted befre users are allwed t perate the centrifuges. 2 Operating and maintaining all centrifuges in accrdance with manufacturer s instructins and gd safe labratry prcedures. Reading and fllwing all instructins fr safe usage and maintenance f the centrifuge. Selecting the apprpriate rtrs/tubes fr the prtcl in use. NOTE: IF AT ANY TIME YOU ARE UNSURE HOW TO OPERATE THE CENTRIFUGES SAFELY, PLEASE REFER TO THE INSTRUCTION MANUAL FOR FURTHER DETAILS AND/OR ASK THE DESIGNATED KGHRI STAFF MEMBER FOR ASSISTANCE. Filling ut the Centrifuge Usage Lg each time the centrifuge is used. Reprting any damage t the centrifuges r the rtrs t the designated KGHRI staff member immediately s equipment repairs can be made. Wearing apprpriate PPE (lab cat, safety glasses, and glves). KGHRI is respnsible fr: Ensuring all centrifuges are in gd repair and labelled with apprpriate cntact infrmatin fr maintenance and repair. Training new users in safe usage and maintenance f the centrifuges, dcumenting the training f all users, and supervising use when necessary. Ensuring that the Centrifuge Usage Lg is kept clse t the equipment and that all users fill ut the Centrifuge Usage Lg. Ensuring all cpies f user instructins n balancing lads in the rtr, maximum G lads in RPM, and filling and sealing samples are readily available t all users. Ensuring that all the regular and peridic maintenance required is carried ut and recrded in the Maintenance Lg. Ensuring the recrds f repair are kept and available fr inspectin. 3.0 PROCEDURES Operatinal Prcedure Rtr Buckets (metal) and Rtr Bucket Inserts (sample tube hlders) It is imprtant t always use the crrect rtr bucket insert fr the tubes t be centrifuged. Rtr bucket inserts are always used in PAIRS and MUST ALWAYS ccupy the bucket directly acrss frm each ther. Versin 1 (December 1, 2017) - 2 SOP-CS-01

3 Lading f Samples Lad tubes int the buckets. Use the aersl resistant caps prvided fr the rtr buckets. Clse the rund bucket using an aersl-tight cap. Check the sealing ring (O-ring) in the cap (Item 1 in the figure belw) t ensure that the sealing ring is nt damaged and is seated evenly in the grve. Place the aersl resistant caps n and screw the caps n tightly. Symmetrically distribute balanced buckets r tubes. Always use tubes f the same type (weight, material/density and vlume). Always perate the centrifuge with all buckets in place, even if tw ppsing buckets are empty (see image belw). RUN ROTORS WITH A FULL SET OF BUCKETS. Versin 1 (December 1, 2017) - 3 SOP-CS-01

4 Instrument Safety Features All centrifuges must have a lid lift assisting struts which enable the lid t be raised withut effrt and hld the lid in the upper mst psitin fr lading, unlading and servicing. IMPORTANT: As the lid lift assist struts age they slwly lse the ability t hld the weight f the lid. When this situatin is identified, ntify the designated KGHRI staff member as a service call must be placed t crrect this safety issue. All centrifuge lids must be lwered t the clsing psitin and then slight pressure needs t be applied t the frnt edges f the lid until the electrnic lcking mechanism engages. This will be an audible clicking sund. NOTE: The centrifuge will nt perate unless the electrnic lcking mechanism is engaged. If the instruments lid fails t lck ntify the designated KGHRI staff member as a service call must be placed t crrect the issue. Pressing the start/stp buttn at any time during the centrifuge cycle will cause the centrifuge t stp the rtr. Autmatic braking will ccur under nrmal cnditins f lad imbalance. Emergency Lid Unlck IMPORTANT: The centrifuge must be unplugged befre starting this prcedure. The rtr must nt be turning when trying t pen the lid. Wait until the rtr has stpped turning (t check, lk thrugh the mnitring glass in the centrifuge lid) befre prceeding with the emergency unlck prcedure. Each centrifuge will have a different emergency lid unlck methd. Cnsult the individual peratr s manual fr specific details. Initiating Centrifugatin Cycle Prir t pressing the start buttn ensure the fllwing has ccurred: The centrifuge is switched n. The rtr has been inserted and attached crrectly. The rtr has been laded crrectly. Buckets can freely swing ut. The centrifuge lid is clsed. The crrect prgram r speed/run duratin time/acceleratin-deceleratin has been selected. Setting the Centrifugatin Parameters Centrifugatin Time (min:sec): use the rtary knb TIME t set the centrifugatin time. Versin 1 (December 1, 2017) - 4 SOP-CS-01

5 Refrigerated Centrifugatin Temperature (-9 C t 40 C): When the centrifuge pwer is n, the default temperature is set at 4 C. T change the temperature, use the TEMP arrw keys t set the temperature. Centrifuge Speed: Use the rtary knb SPEED t set the speed f the centrifugatin. Maximum speed: 3000 x g (4400 rpm). Press knb briefly t switch display f centrifugatin speed (rpm r rcf). Start the centrifuge cycle. D nt leave the centrifuge until full perating speed is attained, and the instrument appears t be running nrmally withut vibratin. Nte: It is nrmal t see a small vibratin in the centrifuge as the speed increases, but any large vibratin in the centrifuge r irregular nise may indicate that smething has gne wrng with the run. Press the STOP buttn and make sure that the tubes are balanced prperly. If centrifuge cycle is successful, wait until the centrifuge cmes t a cmplete stp prir t pening up the unit. Open the centrifuge carefully. Remve the sealed buckets (r tubes) slwly and carefully t prevent re-suspensin f the sediments; always check fr damage t yur tubes. Sediments and supernatants shuld be visible after centrifugatin. When using the refrigerated centrifuge, the centrifuge lid shuld be left pen r ajar when the centrifuge is turned ff at the end f yur specimen prcessing s that any cndensatin present dries as quickly as pssible. Errr Cdes Each centrifuge has prgrammed errr cdes t assist in identifying the prblem. Cnsult the individual peratrs manual fr specific details (lcated in the binder n the shelf abve the centrifuges). If the centrifuge stps running r will nt start, write dwn the errr cde frm the main display, and ntify the designated KGHRI staff member as a service call might be required t crrect the issue. Reference Manuals The Instructin Manual fr the ambient and refrigerated centrifuges (same manual) is lcated in the research centrifuge rm (Rm ) in the binder n the shelf abve the centrifuges (Eppendrf Centrifuge 5702/5702 R/5702 RH Operating Manual). 6 4.0 MAINTENANCE Device The uter surfaces f the centrifuge and the rtr chamber shuld be cleaned regularly with a mild cleaning agent. This is fr hygiene purpses and t prevent adhering impurities causing crrsin. Befre cleaning, unplug the pwer plug with the lid pen, unscrew the rtr using the rtr key supplied and clean it separately. Use Oxivir wipes fr cleaning the accessible surfaces f the equipment. D nt allw any liquid t get int the gap at the mtr shaft utlet. Fr this reasn, the rtr chamber shuld be cleaned nly with a damp clth. The rubber seals in the rtr chamber shuld be rinsed ff thrughly with water and lubricated with glycerin (supplied) after every clean t prevent them becming brittle. Rtrs The rtrs, buckets, tube hlders, caps, and adaptrs shuld be cleaned nce a mnth r when necessary with a neutral cleaning agent t prevent residues f the fluids being centrifuged frm changing the prperties f the centrifuge and its accessries. The rtr must be taken ff fr this. Check the tube hlders and buckets fr residues and crrsin. Fr thrugh cleaning, remve the rubber plates frm tube hlders and buckets and clean all parts separately. Then refit the rtr and blt it int place with the supplied rtr key. Check the rtr, tube hlders, caps and buckets nce a mnth fr mechanical damage. The aersl-tight cap, including the silicn ring, rubber mats and bucket adaptrs are subject t nrmal wear and must be replaced when a visual inspectin reveals wear. On the swing-bucket rtr make sure, in particular, that the pegs and grves f the buckets are free f dirt. They shuld be lightly lubricated with pivt grease (supplied) s the buckets can swing freely. The aersl-tight buckets MUST NOT be stred with their caps sealed. Refrigerated Centrifuge Clean cndensatin water and ice buildup regularly frm the rtr chamber (by defrsting), using a sft, absrbent twel. Regularly empty and clean the cndensatin water cllectr. Remve this frm the left. Please clean the cndensatin water drain regularly. 6.0 SPILLS IN A CENTRIFUGE When yu becme aware f a spill in a centrifuge, infrm any nearby ccupants that a spill and likely tube breakage has ccurred inside the centrifuge. Pst a Bihazard Spill sign ON the centrifuge (lcated in the cabinet under the dirty sink): Versin 1 (December 1, 2017) - 6 SOP-CS-01

7 Ensure the lid remains clsed; d nt disturb the centrifuge fr 30 minutes after the rtr has stpped t allw aersls t settle. PPE MUST be wrn befre prceeding t the next step. Obtain Oxivir slutin and Oxivir wipes lcated in the cabinet under the dirty sink. Open centrifuge carefully t avid disturbing the cntents which may be up against the lid. Assess the extent f the spill and damage f the tubes, buckets, rtr and inner surfaces f the centrifuge. IF YOU ARE ALONE AND NOT ABLE TO CLEAN UP THE SPILL, cntact yur supervisr/manager and the designated KGHRI staff member. Remve any debris using frceps, tweezers r tngs and place in the yellw Sharps waste bin/cntainer. Carefully remve rtrs and buckets and place in the plastic centrifuge tub lcated under the dirty sink. Mve the centrifuge tub t the bilgical safety cabinet (BSC) fr further cleaning. Pur cntents f disinfectant (Oxivir ) int centrifuge tub allwing the rtrs and buckets t sak in the BSC. Ensure all reusable items used in the cleanup (e.g. frceps, dustpans, etc.) are als saked in disinfectant (Oxivir ). After the recmmended cntact time with Oxivir slutin (5 minutes), remve the centrifuge tub frm BSC and place cntents int dirty sink. Rinse cntents thrughly with water in the dirty sink. Wash cntents with sap and water using cleaning tub lcated in the cabinet under the dirty sink. Rinse cntents and lay ut t dry using paper twels n bench. Return cntents t the centrifuge r strage bin when dry. Dispse spent disinfectant (Oxivir ) frm centrifuge tub and sapy water frm cleaning tub dwn the dirty sink with running water. Wipe the inside f the centrifuge with disinfectant (Oxivir ). Take steps t inactivate any cntaminatin n surfaces befre tuching the surfaces. If needed, use paper twel dampened with disinfectant (Oxivir ) t clean the inside f the centrifuge. All used paper twels saked in Oxivir and/r Oxivir wipes are t be discarded in the yellw bihazard waste bin/cntainer. Remember t wipe ut the BSC with Oxivir wipes as per nrmal cleaning prcedures utlined in Bilgical Safety Cabinet SOP. Place all ther waste (e.g. paper twels used t dry centrifuge parts that were nt saked in Oxivir ) int regular waste bin/cntainer.
